Opener-wicketkeeper Juyal (201 off 278 balls, 21 fours, 1 six) and Sharma (208 off 310 balls, 23 fours, 3 sixes) amassed a 298-run partnership for the third wicket while notching up their maiden first-class double century.
Akshdeep Nath scored a half-century down the order to help UP rack up a huge total.
For visitors Assam, medium pacer Mrinmoy Dutta (4/116) was the most successful bowler while left-arm orthodox spinner Rahul Singh returned 3/142.
In reply, Assam had a strong start, reaching 116 without losing any wicket at stumps with openers Parvej Musaraf (53 batting) and Rahul Hazarika (51 batting) remaining unbeaten playing similar kind of innings. However, on Day 3, the Riyan Parag-led visitors have a mountain to climb as they trail UP by 432 runs.
