Jyothirmayi started her cinema journey at the age of 17 and played a supporting role in the Malayalam film 'Pilots' starring
Suresh Gopi in the lead role. Jyothirmayi then continued to stay with Malayalam for five years and made her Tamil debut with the 2005 'Sivalingam IPS'. But it couldn't help her to gain fame among the Tamil audiences.

Jyothirmayi then played the female lead in Sundar C starrer 'Thalai Nagaram', which was directed by Suraj, in 2006. Jyothirmayi played a young model, who falls in love with a gangster played by Sundar C, and she had an extended screen time. The massive success of the crime action transformed her into a popular actress, and she bagged back-to-back Tamil films.
Jyothirmayi's role in 'Thalai Nagaram' is an ever memorable one, then she opted to do challenging roles, and placed herself in notable films. Jyothirmayi later made her Kannada debut and developed into a notable South actress. 'Sabari', 'Naan Avan Illai', 'Periyar', 'Arai En 305-il Kadavul', and 'Vedigundu Murugesan' are other notable films of the actress in Tamil. But she kept her more active in Malayalam and delivered more films in her native language.

When we look into the personal life of Jyothirmayi, the actress married Nishanth Kumar in 2004, but their relationship didn't last longer as they got divorced in 2011. Jyothirmayi then married Malayalam director and cinematographer Amal Neerad in 2015, and she is off from films since 2013. It is worth mentioning that Jyothirmayi made a special appearance for a song in the Malayalam film 'Sagar Alias Jacky Reloaded', which was directed by Amal Neerad.
